The consortium is composed of a multidisciplinary group of basic, translational, and clinical scientists from a broad range of disciplines who share a commitment to develop programs that will improve the survival of patients with gastrointestinal cancer. The University of South Florida/St. Petersburg has an established record in contributing to clinical and basic cancer research. A new Florida SPORE in Gastrointestinal Cancer addresses three important translational problems in gastrointestinal cancer: Detection, tumor progression, and therapy and control. This application has two projects and two cores. Project 1, entitled, “High-Throughput Analysis of Omic Profiles for Tumor Classification,” has as its central theme differential expression of key biochemical pathways, referred to here as omic profiles, as a means of discriminating between normal and malignant tissues. This research will involve data mining the omic profiles of normal and malignant gastrointestinal tissue samples that will be generated by the Genomics and Bioinformatics Core (Core B). The Omic Profiles Core will provide a platform for data deposition, assessment, and mining to develop differential expression signatures to be used in Projects 2 and 3 for the purpose of detection and classification of gastrointestinal tumors. Project 2 will study the expression patterns of a broad range of genes, including cancer/testis antigens, secreted factors, and apoptosis related genes, in intraepithelial neoplasms, villous adenomas, and adenocarcinomas in order to define the molecular pathways responsible for tumor progression. In Project 3, the role of specific protein kinase pathways in the progression of gastric and colon cancers to highly aggressive, metastatic disease will be assessed. This project will involve mouse transgenes with specific alterations of the mitogen activated protein kinases and the phosphoinositide 3-kinase pathways, and human xenograft studies.
