By applying current recombinant DNA techniques, the investigator will express and characterize wild type and mutant factor V proteins in both human and baculovirus expression systems. The investigator’s previous work on these mutations suggest both a correction of the factor V-deficient phenotype through restoration of the cofactor activity and a treatment through the inhibition of the proteolytic cleavage of factor V. The investigator will also probe the novel mutations for the presence of cofactor activity and to assay their effect on proteolytic processing of the factor V in the presence of factor Xa. In the last section of the proposal, the investigator will use these studies to develop novel therapeutic modalities against the novel factor V mutations by creating targeted inhibitors. The investigator will target the novel mutations with both small molecule and peptide probes as a long-term goal. The investigator will also develop efficient DNA-based methods for treatment of these factor V-deficient patients. PROPOSED COMMERCIAL APPLICATION: In the U.S. alone there are probably at least 100,000 genetically hemophilia and 300,000 VWD cases which are not dealt with by present therapies. The availability of targeted therapy to these patients would be a revolution in the management of these disorders.
